Highlights
We discovered a mechanism by virtue of which the transcription factor Alx3 downrrgulates the expression of glucagon in pancreatic islets in response to elevations in the concentrations of glucose. This mechanism is important for the maintenance of glucose homeostasis, and may have implications in
the etiopathogenesis of diabetes, in patients showing paradoxically hyperglucagonemia accompanying hyperglycemia. Notably, the mechanism of action of Alx3 at the molecular level appears to be conserved in other systems, for example in the inhibition by Alx3 of the expression of genes that are important for the differentiation of melanocytes in the skin.
